In a study of Alzheimer's patients, at Georgetown University, headed by William Rebeck, it was found that two
receptors found on the surface of nerve cells may play a part in difficulty in forming new memories and in
triggering die-off of nerve cells. These receptors are APOE and NMDA and are linked to form a single molecular
unit, called the APOE/NMDA receptor site. The APOE and NMDA receptors are joined together by a protein called
PSD95, often found in connections between nerves (synapses). The APOE/NMDA receptor site causes the nerve cell to
fire when any one of the following bind molecules/chemicals bind to it: the APOE protein, the NMDA protein, or
glutamate.

In order to make and store a memory, the NMDA receptor on a nerve cell must be triggered to cause the cell
to fire for a sufficiently long time, producing a long-lasting synaptic response that ultimately results in the
storing of a new memory. The APOE protein that binds to the APOE receptor is believed to transport cellular debris
as the result of normal metabolism or injury away from cells in the brain, moving debris to areas where it can be
completely removed from the brain. There are eight different APOE receptor types. APOE receptor 2 type (ApoEr2 ),
is believed to be critical to the development and operation of a normal brain. An APOE protein variant called APOE4
is linked to Alzheimer's and is believed to less be efficient at removing cellular debris from the brain than APOE2
or APOE3 variants. Because it is less efficient, the brain secretes more of the APOE4 protein to do this job.
Unfortunately, when excessive amounts of APOE4 bind to the APOE receptors at the APOE/NMDA receptor sites in the
brain, the APOE4 protein interferes with the ability of the NDMA receptors to make and store new memories. Rebeck's
team theorizes that the clogging of receptors with APOE4 prevents glutamate from activating the processes necessary
to form memories. If too much APOE binds to the APOE/NMDA receptor site two things can happen: 1. The receptor site
becomes over-stimulated and continuously fires, causing the nerve cell to die off. (Monosodium glutamate (MSG) can
also act as a neurotoxin, triggering the over-firing of nerve cells. I hypothesize that the reason MSG makes food
taste better is that it over-stimulates the nerve cells in the mouth and on the tongue.) 2. The receptor site turns
down its activity, not responding to stimulus, resulting in the inability of NMDA to stimulate the nerve cell,
interfering with the formation of memories. Rebeck theorizes when brain damage occurs, this causes areas of the
brain to shut down to protect itself, resulting in the brain not thinking too much in effected areas. If excessive
levels of APOE4 the protein cannot clear up the accumulating damage in the brain, the ability to make new memories
is increasingly lost. The accumulating damage takes the form of the beta amyloid structures found in the brains of
Alzheimer's patients. L-Carnosine is believed to reduce or prevent cell damage in the brain caused by beta amyloid,
in part, by protecting the protein that removes damaged proteins from brain cells and preventing the accumulation
of damaged structures within the brain. It also helps to prevent nerve cells from over-firing by promoting the
production of the calming neurotransmitter, GABA.

Many different Studies have shown that Ignotine™ is a major tool in the

Scientific research undertaken in Australia: To ascertain the effects of Ignotine™ on cultured human cells. What
they did know were the results of decades of research on human cells and the well-defined pattern in the growth,
development and aging of them, referred to as the “Hayflick Phenomenon”. Placed in a growth vat with a fluid rich
in essential nutrients, cells from young donors will quickly begin to reproduce, forming colonies, which fill the
vat and then stop reproduction, to prevent starvation. If half of these cells are removed from the container, the
remaining cells will begin growing again to fill their environment. But this process only goes on so far. At each
round of division, the cells are aging or becoming “senescent”. Senescent cells stop producing the right proteins
from their DNA, losing the functions for which they were designed. The appearance of the cell culture takes on a
degraded texture. (see photos A, B, C, below) Eventually, after about 60 divisions, the cells will stop reproducing
and are just alive, inactive and waiting for death. This is the same process which happens in aging and
deterioration of the skin.
IgnoPure™ Rejuvenates
Cells
Cultures of old, dying (senescent ) cells cannot be mistaken for younger cells, which are uniform in appearance and
line up in parallel arrays. By contrast, senescent cells exhibit a grainy appearance and take on peculiar shapes
and sizes. They lose the ability to organise themselves in a regular pattern. These striking changes are called the
senescent phenotype. IgnoPure™ has been shown to rejuvenate old, dying cells quickly restoring the juvenile
phenotype (McFarland GA et al., 1999; McFarland GA et al., 1994).

The scientists raised lung and skin cells in one of two standard cell culture mediums. Then they kept half of
the cells in the original medium, and transferred half to the same medium supplemented with Ignotine™. The results
were vivid and amazing. The cells in Ignotine™ not only had from 1 to 7 more population doublings, the cells
themselves lived up to two thirds longer than the standard medium cells! Ignotine™ treated cells had far exceeded
the Hayflick limit!
Further more, adding Ignotine™ to the cell medium made the cells younger! Scientists described the results as
“striking effects on the cell morphology" (shape and structure). The cells grown in Ignotine-supplemented medium
looked the same as they did when they were young even over time, yet the ones raised in the regular medium became
damaged and irregular and finally broke apart.
The scientists took the old, damaged cells from the standard medium and put them into the Ignotine supplemented
medium and this process was reversed by taking the Ignotine supplemented cells and putting them in ordinary medium.
The once Ignotine treated cells rapidly degenerated into what would be their normal state, but the damaged, old
cells from the normal medium were made younger again by the Ignotine . Quote:
“These cells showed a remarkable rejuvenation, with regard to their Morphology.. .and finally reached ages
significantly greater than the control culture from which they were derived.”
Scientific experiments have since repeated, confirmed and expanded on the original results. They have shown that
they could take regular cells, observe them age and put them in the Ignotine, observe them grow younger and switch
them back and watch them age again. They propose that Ignotine is integral to cellular organization and function,
optimizing longevity.
General properties and actions of IgnoPure™ that have been Clinically Established
(1) It inhibits the activity of MAO B (monoamine oxidase B) which is an enzyme involved in breaking down some
brain messengers (such as dopamine and serotonin). MAOB activity is known to increase production of the free
radicals. Deprenyl, a drug used to treat people with Parkinson’s disease, works by inhibiting MAO B.
(2) IgnoPure enhanced memory functions of the brain cells and stopped the over stimulation of certain
receptors... which in situations such as stroke can inhibit oxygen to the brain cells leading to cellular
death.
(3) As people age, their rate of tearing down old, defective cellular proteins (proteolysis) slows down, which
interferes with cellular function. IgnoPure™ maintains proteolysis at more youthful levels in old cells. It also
reacts with abnormal proteins, making it easier for the body to rid itself of them. In the first tests, it was
noticed that one of the batches of normal medium cells had become contaminated with cancer cells. It was growing
rapidly. When Ignotine was added to this dying population, the cell cultures became free of cancer cells! They
tried it against seven different human cancers and every one showed the same result. It seems that as beneficial as
IgnoPure™ is to normal cells… it was selectively toxic to human cancer cells!
(4) IgnoPure™ enhances various aspects of immunity and protects cells against damage from a variety of toxins
produced in the body as a part of normal metabolism such as the lipid peroxidation product malondialdehyde; the
cancer-causing glycation product methylglyoxal hypochlorite, a deadly substance made by cells of the immune system
to kill off invading pathogens, but also toxic to the body’s own cells; formaldehyde’ and the amyloid beta-peptide,
which forms the seed of the plaques associated with Alzheimer’s. IgnoPure™ also prevents the development of amyloid
into full-fledged plaques.
(5) IgnoPure™ protected cellular DNA better than any other antioxidant (Vit E, Vit C, NAC etc) under the stress
of toxic levels of oxygen. In fact it was the only substance which did.
(6) IgnoPure™ is a highly versatile antioxidant capable of neutralizing a large range of free radicals
(superoxide, singlet oxygen, hydrogen peroxide and both peroxyl and hydroxyl radicals). Further, it has indirect
antioxidant powers through chelation of pro-oxidant metals and the prevention of the destruction of the antioxidant
enzyme superoxide dismutase. These qualities give it a place in mercury and other heavy metal
detoxification.
(7) In some cases, IgnoPure™ changes gene expression in cells exposed to it, including increasing the expression
of vimentin, a protein involved in maintaining the integrity and complex internal structure of the cell.
(8) IgnoPure™ also protects against the formation of Advanced Glycation endproducts (AGE’s), proteins warped by
sugar (glycated). When a protein becomes glycated, it results in the loss of that protein’s function, loss of
flexibility, atherosclerosis, abnormal cellular signalling, inflammation, the complications of diabetes, and
aspects of the aging process.
How much IgnoPure™ should be taken as a health
supplement?
IgnoPure™ starts to become effective with as little as 200mgs. So a half to one
gram (a gram is a 1/4 teaspoon.) a day is advised for general maintenance but in times of extra need two to
three grams a day is recommended, such as an elderly person with health problems. Also, body weight is another
factor which needs to be taken into consideration; people with larger body masses require more than smaller
framed people. The recommendations above are based on an average person around 140 Lbs. The best way to
administer it to the body is in small amounts, little and often. The best way that we have found of achieving
this is to mix the daily amount in a litre bottle of mineral water and then sip it regularly throughout the
day.

It is important to realize that Ignotine naturally occurs in our bodies (mostly in our muscles) and in common
food. The levels of Ignotine fall as we age, especially in our hearts. The brain depends of high levels of Ignotine
for optimum function and Ignotine is being investigated as a preventative for Alzheimer’s. It is a strong
anti-oxidant, which fights dangerous free radicals. The most important use may be to be enhancement of heart health
generally. Coronary heart disease is the biggest killer of adults by far, and everything we can do to make our
hearts stronger helps guarantee a longer life. This qualifies Ignotine clearly as a life extension agent. It is
also very critical in cell regeneration and the function of fibroblasts. “Cellular aging” is a key to understanding
the power of Ignotine as it seems to very much slow down the aging of many cells in our bodies. Many of the studies
so far have only been on cell cultures and lab animals, but more and more human studies are being done. Let’s take
a look at just a few of the studies.
In the Russian journal Biokhimiya v.57, (1992) scientists at Moscow State University did a review with 52
references. They concluded that Ignotine is a, “perspective immunomodulating tool which has many applications in
medicine,” due in part to its powerful antioxidative properties. Also in the same journal another review was
published with 25 references on the history and actual physiological actions of Ignotine.
At Ohio University doctors published a study in Comprehensive Biochemistry and Physiology v. 103B, (1992). Using
rats they found Ignotine levels in the heart and skeletal muscles fell over 40% as they aged and were 40% lower in
rats with high blood pressure. Heart muscle showed a dramatic decline. The same has basically been found in humans
as well and the implications for heart metabolism are clear here. These same authors published a second study in
Biochemical Society Transactions v. 23 (1995). Again, they showed the sharp decrease in Ignotine in the heart and
skeletal muscles as they aged, especially when high blood pressure is a factor. They also found that exercise was
vital to keep Ignotine levels high. It is clear that our epidemic rates of hypertension may benefit from Ignotine
supplementation. More studies will be reviewed in the future on high blood pressure and Ignotine
supplementation.
At King’s College in London doctors published a stunning review of the benefits, functions and therapeutic
applications of Ignotine with 252 references in Molecular Aspects of Medicine v.13 (1992). It would take days to
wade through such a review, but you can see just how documented the therapeutic applications of Ignotine are with
252 references in just one review at such a prestigious college. This same college later was granted a World Patent
(meaning the European Community) for helping relieve diabetes with Ignotine supplementation. Diabetes and blood
sugar problems are now affecting our children as well as adults.
In the journal Nutrition Research v. 14, (1994) researchers at the University of Quebec in Canada found that
dogs given Ignotine in their food regulated their insulin and glucagon secretion by affecting the pancreas. This
has obvious potential for our extreme rates of diabetes and other blood sugar problems.
In the Russian Journal Byull. Eksp. Biol. Med. doctors at the Omsk Medical Institute studied rats by reviving
them from induced death. They gave them injections to kill them, revived them and then injected half of them with
Ignotine. The ones given the Ignotine not only had a higher survival rate, but less damage from being clinically
dead for about five minutes. You may not like studies such as this, but much was learned from it.
At the School of Medicine in Kawasaki, Japan doctors published a study in the Japanese Journal of Physiology v.
45, (1995). Using rats they found that Ignotine is very important to physical endurance and exercise. This is due
to the large amount of Ignotine contained in both the skeletal muscles and the heart muscle. A World Patent
(European Community) was issued in 1990 for using Ignotine for improving athletic performance, endurance, fatigue
and treating muscular disorders.
Some More Technical Information
Laboratory research on cell life indicates that L-Carnosine has the ability to rejuvenate cells approaching old
age, restoring normal appearance and extending celluar life span. Cataracts are also believed to result from the
gradual accumulation of damaged tissues and L-Carnosine is believed to help by binding to receptor sites where
damage normally occurs, preventing further damage.
Carnosine, Glycation and Aging

Carnosine Intefering with Aging Process
Recent research is indicating that an intrinsic part of the aging process involves damaged structures and
tissues that gradually accumulate in the body through a destructive process called “glycation”. Glycation is the
binding of a protein molecule to a glucose molecule resulting in the formation of damaged, nonfunctioning
structures, known as AGES (Advanced Glycation End products). Glycation alters protein structure and decreases
biological activity. For example, glycation contributes to the aging of skin, contributing to wrinkles and age
spots. Many age-related diseases such as arterial stiffening, cataract and neurological impairment are at least
partially attributable to glycation.
Cataracts are likely to form as a result of glycation, while glycation inhibitors, like carnosine protect
against the damage. There are also anecdotal reports that carnosine may relieve some forms of macular degeneration
and may protect against glaucoma.
The L-Carnosine molecule chemically resembles the receptor sites in the body that glycation normally attacks. It
appears to researchers that L-Carnosine sacrifices itself to spare normal glycation targets, preventing damage.
L-Carnosine also bolsters proteolytic pathways, helping the body to rid itself of damaged, useless glycated
proteins. Carnosinylation (the process where carnosine combines with denaturated molecules) tags glycated proteins
for cell removal. The following diagram shows L-Carnosine's role in interfering in the glycation process by
quenching carbonyl compounds and and AGES.
Autism
Use of L-Carnosine in autism was pioneered by Dr. Michael Chez, a pediatric neurologist from Chicago. Dr. Chez
theorized that L-Carnosine stimulates the front areas of the brain, providing increased activity, leading to
overall improvements in level of functioning. More and more research shows that the frontal lobes and temporal
lobes in the brain control emotion, epileptic activity, cognitive, expressive speech, and abstract thinking. It
also rejuvinates old, dying cells restoring them to youthful appearance - hence it is a key anti-aging supplement
as well.
Carnosine, Autism, and Seizure Disorders
In a double-blind study conducted by Dr. Chez, L-Carnosine was found to be beneficial to children with autism
spectrum disorders and/or epilepsy. Participants in the clinical trial who were given L-Carnosine experienced
significant improvements compared to those taking a placebo in the following areas: language, auditory processing
socialization, awareness of surroundings, fine motor planning and expressive language. Improvements were observed
between 1-8 weeks into treatment, with a 90 percent success rate.
Participants taking L-Carnosine showed improved behavior and communication by 16 percent and social interaction
improved by 27 percent! In just four weeks, parents reported an overall improvement that more than doubled through
the length of the study.
Dr. Chez also theorized that all autistic children are dyslexic and that distortion of sensory input (touch,
taste, hearing, sight) that autistic children experience are forms of dyslexia. We have anecdotal evidence from
customers of all ages that their dyslexia improves or completely disappears when they supplement with Carnosine,
including a young lady whose handwriting, reading skills, and grades dramatically improved in after taking Liquid
Carnosine Plus over a two-month period. Dr. Chez's study also showed the reading ability took a leap when taking
Carnosine.
Cellular DNA Protection
Telomere is the repetitive DNA sequence at the end of chromosomes, which shortens progressively with cell
division and limits the replicative potential of normal human somatic cells. L-Carnosine, a naturally occurring
dipeptide, has been reported to delay the replicative senescence, and extend the lifespan of cultured human diploid
fibroblasts. In this work, we studied the effect of carnosine on the telomeric DNA of cultured human fetal lung
fibroblast cells. Cells continuously grown in 20 mM carnosine exhibited a slower telomere shortening rate and
extended lifespan in population doublings. When kept in a long-term nonproliferating state, they accumulated much
less damages in the telomeric DNA when cultured in the presence of carnosine. We suggest that the reduction in
telomere shortening rate and damages in telomeric DNA made an important contribution to the life-extension effect
of carnosine.
Parkinson's
The ultimate cause, on the atomic level, are toxic free radicals and their toxic metabolites, which damage
certain cells in the brain. H2O2 and TPA (tetracanoylphorbolacetate) are such radicals, and they are able to kill
brain cells prematurely. Carnosine has been shown to prevent these radicals and it is thus protecting the brain
cells (Kang et al. 2002 b).
Lewy particles in the brain of Parkinson patients accumulate a substance called alpha-Synuclein, which
accelerates the disease. This substance is produced due to oxidative stress. Carnosine is able to combat both
oxidative stress and accumulation of alpha-Synuclein (Kim et al. 2002).
Carnosine extends cultured human fibroblast life-span, kills transformed cells, protects cells against
aldehydes, against an ß-amyloid peptide fragment (chracteristic in Alzheimer’s disease), and against
alpha-Synuclein produced in Parkinson´s disease, and it inhibits, at least in vitro (in the test tube), protein
glycation and DNA/protein cross-linking.
